Transcriptional Targets

Were the binding sites shown derived from experimental results? Or were they from computational methods?
The data presented on the p53 target genes were obtained through manual curation of available papers, as well as existing databases such as BIND, TRANSFAC and Entrez Gene. So the binding sites shown on the p53 knowledgebase were obtained from published experimental results and not based on any binding site predictions.

Mutations

Why are the mutation frequencies shown on the p53 knowledgebase different from those generated by IARC?
Mutation frequencies obtained from the p53 knowledgebase differs from the ones obtained from IARC database search system because of the following reasons:
  • While IARC stores the start positions of deletions, the p53 knowledgebase stores information on block deletions with start and end nucleotide positions. Hence, the mutation frequencies inlcude deletions that are found within these deletion blocks, resulting in higher number of selected deletions than in the IARC database search system. Please refer to the curation process for details.
